- Troubleshooting the chromatogram through an evaluation of;
* The separation chemistry - pH and buffer strength effects
* Changes to the chromatographic baseline – cyclical, fluctuating etc.
* Changes to chromatographic peak shape – tailing, splitting etc.
* Changes in the HPLC system operating pressure and analyte retention times and peak areas
